Background
Ibutamoren (MK-677) was designed to treat growth hormone deficiencies which may occur during childhood or as an elder. A deficiency of this hormone results in frailness and/ or stunted growth. By acting as a ghrelin mimetic, Ibutamoren stimulates both appetite and muscle fiber synthesis, normally resulting in weight gain by subjects given it. Pharmacology
Ibutamoren (MK-677) is a potent, long-acting, orally-active, selective small molecule agonist of the ghrelin receptor and a growth hormone secretagogue, mimicking the growth hormone-stimulating action of the endogenous hormone ghrelin. It has been shown to increase the secretion of several hormones including GH and insulin-like growth factor 1 (IGF-1) and produces sustained increases in the plasma levels of these hormones.
The effect of MK-677 on prolactin is variable, with adolescents experiencing no change[1] and elders experiencing a 27% increase.[2] The same inconsistency is shown with cortisol, wherein one study shows an increase[3] and another shows no change.[4]
